Dr. Sheldon Cooper, theoretical physicist
Cooper works at Caltech and is known for his brilliant mind and quirky behavior. In the early stages of the show, Cooper is shown to be uncomfortable with relationships and intimacy; however, he ends up falling in love and eventually marrying neurobiologist Amy Farrah Fowler, with whom he now lives. He previously lived with his best friend, Leonard Hofstader.

Penny, pharmaceutical sales rep (former waitress)
Penny moved from a small town of Nebraska to California with the dreams of making it big in Hollywood. She worked at the Cheesecake Factory as a waitress and after dating Hofstader on and off for a few years, she finally marries him. She did not become an actress but is now a successful sales rep.

Rajesh Koothrappali, astrophysicist
Raj is an astrophysicist at Caltech and also presents shows at the Griffith Planetarium. Friends with Sheldon and Leonard, he is best known for his inability to talk to women without being inebriated. He has had his fair share of love interests in the show, but remains out of luck when it comes to settling down.
Howard Wolowitz, aerospace engineer (former NASA astronaut)
Wolowitz also works at Caltech and is best friends with Koothrappali. Wolowitz went from trying to be a "ladies' man" in the early seasons of the show to being the first guy in the group to get married and settle down. He is married to Bernadette, and the two of them have a son and a daughter together.
Amy Farrah Fowler, neurobiologist

Fowler joined the group in season three of the show, and is a neurobiologist. She meets Sheldon after Wolowitz and Raj create a fake dating profile for Cooper and she matches with him. She is often deemed to be quite similar to Sheldon and the two end up falling in love and getting married. Amy considers Penny as her best friend.
Bernadette Rostenkowski, microbiologist
A microbiologist, Bernadette joins the show in season three. She begins dating Wolowitz and the two get married in season five. She is perhaps best known for her high-pitched voice, which sounds eerily similar to Wolowitz's mother's, when she is angry.
Stuart Bloom, owner of The Comic Center, a comic book store
Stuart is the owner of the comic store that the guys are often seen visiting. He first appeared in the show in season two, has often tried to be a part of the group and has expressed romantic interest in both Penny and Amy. He currently has feelings for Denise, whom he hires as an assistant manager for his store.
Will Wheaton as himself (actor, blogger, writer)
Wheaton plays himself in the series and appeared in season three for the first time. He is best known for his love-hate relationship with Sheldon. He finally makes up with Cooper in season five and has, since then, remained good friends with the characters of the show.
